The gene coding for green fluorescent protein (GFP) from the jellyfish Aequorea victoria was successfully used as a vital marker for the transformation of three woody plant species, black spruce (Picea mariana (Mill.) BSP), white pine (Pinus strobus L.) and poplar (Populus spp.). The gfp gene and the gene conferring resistance to kanamycin (nptII) were introduced by microprojectile bombardment or Agrobacterium tumefaciens-mediated technology. Screening by fluorescence microscopy of the transformed plant material, under the selection of kanamycin, identified five to eight cell lines from each tree species that clearly expressed GFP. Expression of GFP was observed in somatic embryonal cells of the coniferous species and in stem sections of poplar. For all species, GFP transgene expression was stable over multiple subcultures. Stable integration of the gfp gene into plant genomes was confirmed by Southern hybridization or polymerase chain reaction (PCR) analysis. We conclude that GFP can be used as a vital marker and reporter protein in transformation experiments with gymnosperms and deciduous trees.  相似文献

Rosebush architecture resulting from the spatial organisation of the plant axes induces plant shape and consequently within ornamental horticulture context, its visual quality and commercial value. This architecture can be modulated by environmental conditions, particularly in the horticulture context in which the possibilities to control growing conditions are numerous. The objectives of the study were to determine, in young rose bushes, (1) whether short periods of nitrogen deficiency affect branching and (2) whether this effect is sufficient to modify the visual quality of the plant in a sustainable manner. Between vegetative bud burst and the petal colour visible stage of the generated primary branch, young rooted cuttings of bush rose (cv Radrazz) were subjected to one of three nitrogen regimes: (1) no nitrogen deficiency, (2) continuous nitrogen deficiency, i.e. 35 days of N deficiency, and (3) nitrogen deficiency restricted to the flowering stages, i.e. 18 days of N deficiency. After the petal colour visible stage, all three groups of plants were supplied continuously with nitrogen. We observed the morphology of the axes and the kinetics of axillary bud burst. Twelve weeks after the petal colour visible stage, the visual quality of the rose bushes was evaluated by an expert jury. We found that nitrogen deficiencies (1) increased bud burst ratios in the medial and basal zones of the primary branch, (2) delayed the bud burst in the apical zone of the primary branch and (3) had long-term effects on plant visual quality. The continuous nitrogen deficiency regime produced flatter, more asymmetric and less vigorous rose bushes than the no nitrogen deficiency regime. By contrast, nitrogen deficiency during the flowering stages only resulted in more symmetric, taller and more vigorous rose bushes than the no nitrogen deficiency regime. Based on these results, the role of nitrogen on bud burst was discussed and candidate processes at the origin of the visual quality modification were suggested. This new approach combining ecophysiology and sensory assessment of ornamental plants enabled the identification of some early architecture components to be correlated with later visual quality characteristics and then to better target the physiological processes of interest.  相似文献

The Interaction between the effects of nitrate (NO3) and sodium chloride (NaCl) concentration on growth) water relations, nitrogen (N) contents and N fixation were investigated in alfalfa (Medicago sativa L. cv. Magali). The plants were grown hydroponically in a growth chamber, in the presence or absence of 3 mM potassium nitrate (KNO3) and exposed to various concentrations of NaCl. Increased salinity resulted in a significant decrease in shoot and root biomass, relative water content and water potential. Shoot growth was more inhibited by NaCl than root biomass. The plants grown in the presence of NO3 were slightly less affected by NaCl than the plants dependent on N fixation for their N nutrition. Nitrogenase activity measured by acetylene reduction activity was substantially inhibited by NaCl, and this inhibition was significantly correlated to the inhibition of shoot growth and total N contents. The comparison of the curves of ARA response to oxygen (O2) partial pressure showed that the salt‐induced inhibition of nitrogenase activity was associated with a significant increase in the critical O2 pressure of the nodules exposed to NaCl. This result shows that NaCl decreases the nodule permeability to O2 diffusion in undeterminate nodule of alfalfa, like previously shown with determinate nodules of soybean.  相似文献

Recurrent or persistent follicular dysplasia and interface dermatitis are described in nine Boxers. Data on age, sex, seasonality of alopecia and histopathological features of the follicular dysplasia in these nine Boxers are comparable with those described in previous reports. The interface dermatitis was characterized by multifocal annular crusted lesions confined to the areas of follicular dysplasia. The inflammatory lesions were neither pruritic nor painful and affected dogs were otherwise healthy. Histopathologically the clinically inflammatory lesions were characterized as an interface dermatitis. Immunohistochemical studies failed to demonstrate immunoglobulins or complement at the basement membrane zone or within blood vessel walls. In dogs with recurrent or persistent disease, the follicular dysplasia and interface dermatitis ran identical, concurrent courses of spontaneous remission and recurrence, or persistence, respectively. One dog with persistent disease was treated successfully with tetracycline and niacinamide for the interface dermatitis, and melatonin for the follicular dysplasia. Although the aetiopathogenesis of this newly described condition and the relationship between the two histological reaction patterns are not known, photoperiod and genetic predisposition appear to play a role.  相似文献

Rice root growth encompasses a remarkable genetic diversity in terms of growth patterns, architecture, and environmental adaptations. In order to harness this valuable diversity for improving rice response to drought, an understanding of key root traits and effective drought response mechanisms is necessary. A trait-based approach with precise understanding of the target environment, including temporal and spatial heterogeneity, is a possible path toward the use of roots and dehydration avoidance traits for improved drought resistance in rice. The ability to grow deep roots is currently the most accepted target trait for improving drought resistance, but genetic variation has been reported for a number of traits that may affect drought response. Here, we review variation in rice root response to drought from a physiological perspective in terms of morphology and function with respect to the different growth environments (upland and lowland) commonly used by farmers. Recent advances in linking physiology and breeding are also presented.  相似文献

A new Integrated Pest Management program based on the combination of synthetic pesticide with a GRAS(generally recognized as safe)-classified sanitizer for the control of Botrytis cinerea in field conditions was described. The aim behind this research was to determine whether the use of this mixture would enhance the efficiency of pesticides while decreasing the recommended dose. Naturally infected tomato plants, grown in the greenhouse, were treated with two commonly used fungicides SWITCH(Syngenta, Switzerland) and SIGNUM(BASF, Germany) each alone or combined with a commercially available organic sanitizer PERACLEAN?5(Evonik Industries, Germany). A total of 27 treatments were tested consisting of three concentrations of synthetic fungicide(×1, ×1/2 and ×1/4 of the recommended dose) either applied separately or combined with three concentrations of the tested sanitizer(0.5, 1 and 1.5%). The control efficacy achieved by the fungicides applied alone ranged between 0 and 66.7% while all fungicide-sanitizer mixtures resulted in up to 70% control of grey mould. The treatment that provides the maximum control of B. cinerea was the result mixture of ×1/4 of the recommended concentration of SWITCH(15 g L~(–1)) with 0.5% of PERACLEAN~?5. This combination suppressed 85% of grey mold infections while decreasing the usually used amount of this pesticide by 75%, reducing therefore the well known negative impacts of chemical pesticides on environment and consumers health.  相似文献

Drought stress is the most important abiotic factor limiting upland rice yields. Identification of quantitative trait loci (QTL) conferring improved drought resistance may facilitate breeding progress. We previously mapped a QTL with a large effect on grain yield under severe drought stress (qtl12.1) in the Vandana/Way Rarem population. In the current paper, we present results from a series of experiments investigating the physiological mechanism(s) by which qtl12.1 affects grain yield under drought conditions. We performed detailed plant water status measurements on a subset of lines having similar crop growth duration but contrasting genotypes at qtl12.1 under field (24 genotypes) and greenhouse (14 genotypes) conditions. The Way Rarem-derived allele of qtl12.1 was confirmed to improve grain yield under drought mainly through a slight improvement (7%) in plant water uptake under water-limited conditions. Such an apparently small increase in water uptake associated with this allele could explain the large effect on yield observed under field conditions. Our results suggest that this improvement of plant water uptake is likely associated with improved root architecture.  相似文献

This work describes betanodavirus infection in two species of groupers (family Serranidae) from the Algerian coast: the dusky grouper Epinephelus marginatus and the golden grouper Epinephelus costae. At necropsy, characteristic clinical signs, external injuries, clouded eyes and brain congestion, generally associated with viral encephalopathy and retinopathy (VER) infection were observed. The partial sequences of RNA1 and RNA2 from two viral strains were obtained, and the phylogenetic analysis revealed the presence of the red-spotted grouper nervous necrosis virus (RGNNV) genotype closely related to strains previously detected in groupers in the same geographic area. Results obtained in this study support the hypothesis that VER disease is endemic in the Algerian grouper population.  相似文献

A new isoquinoline-N-oxide alkaloid was extracted from the alkaloid fraction of a methanol extract of the seeds of Calycotome villosa subsp. intermedia.
